Thyroid stimulating hormone receptor (TSHR) is a guanine nucleotide-binding protein-coupled seven-transmembrane-domain receptor that controls the differentiation, growth, and function of the thyroid gland through stimulation of adenylyl cyclase and phospholipase C pathways. Thyroid stimulating hormone (TSH) is the main TSHR ligand, and unliganded receptor remains silent due to the interaction of its large extracellular domain with the extracellular loops of the serpentine. The TSHR gene is highly mutagenic and constitutively active mutations have been extensively described.

TSH受体(TSHR)是调节甲状腺滤泡细胞分化功能和生长的主要分子。结合TSH后,TSHR通过Gs-cyclic AMP (cAMP)和gq -磷脂酶C级联传导信号。前者调节碘的摄取和甲状腺特异性蛋白的表达,如甲状腺过氧化物酶、甲状腺球蛋白和碘化钠同调体,后者调节碘的外排和H2O2的产生
The TSH receptor is the master regulator of the thyroid. Via the induction of cytosolic second messengers, signaling through the TSH receptor drives the production of thyroid hormones, and the growth of the thyroid follicular cell. These classical activities of TSH receptor activation explain the hyperthyroidism and goiter observed in conditions where there is an abnormal activation of the TSH receptor (such as Graves' disease).
In addition, the TSH receptor is important for the development of the thyroid gland. It is, in fact, expressed during intra-uterine life concurrently with the expression of genes needed for thyroid hormonogenesis, suggesting that TSH/TSH receptor signaling is required for the developing thyroid.
