Designation: | SaOS-2 | Depositor: | Trempe | Organism: | Homo sapiens (human) | Ethnicity: | Caucasian | Age/Stage: | 11 years old | Gender: | female | Tissue: | Bone | Morphology: | Epithelial-like | Celltype: | Osteosarcoma | Growth Properties: | adherent | Description: | The SaOS-2 cell line was established by J. Fogh in 1973. | Culture Medium: | DMEM: Ham’s F12 medium (1:1 mixture) supplemented with 2mM L-glutamine and 5% fetal bovine serum (MG-40, CLS order number 820400). | Subculturing: | Remove medium and rinse the adherent cells using PBS without calcium and magnesium (3-5 ml PBS for T25, 5-10ml for T75 cell culture flasks). Add Accutase (1-2ml per T25, 2.5ml per T75 cell culture flask), the cell sheet must be covered completely. Incubate at 37°C for 10 minutes. Carefully resuspend the cells, the addition of medium is optional but not necessary, and dispense into new flasks which contain fresh medium. | Split Ratio: | A ratio of 1:2 to 1:4 is recommended | Fluid Renewal: | 2 to 3 times per week | Freeze Medium: | CM-1 (CLS order number: 800125, 25ml, 800150, 50ml) | Sterility: | Tests for mycoplasma, bacteria and fungi were negative | Biosafety Level: | 1 | Tumorigenic: | no | Karyotype: | The stemline chromosome number is hypotriploid with the modal number of 56 chromosomes per cell and the 2S component occurring at 13.2%. Over two-thirds of the chromosome complement consisted of structurally rearranged chromosomes. Most marker chromosomes had complex rearrangements. The origin of the segments composing these markers could not be identified. Of the identifiable markers, 6p+/q+, 7p+, 11p+, and 12p+ occasionally were present at 2 copies per cell. The Y chromosome was not detected in the QM stained preparation. | DNA Profile (STR): | Amelogenin: X CSF1PO: 10 D13S317: 12, 13 D16S539: 12, 13 D5S818: 12 D7S820: 8, 10 THO1: 6, 9 TPOX: 8 vWA: 18 D3S1358: 14, 18 D21S11: 28, 30 D18S51: 15 Penta E: 14, 19 Penta D: 11, 12 D8S1179: 10 ,12 FGA: 22, 25 CLS ?
