样品实验方案
简要概述
1.添加 50 μL NaCl 标准品或测试样品
2.添加 50 μL SoNa 520 工作溶液。

4.检测 Ex/Em=490/525 nm 处的荧光
溶液配制
1.储备溶液配制
除非另有说明,否则所有未使用的储备溶液应分成一次性等分试样,并在制备后储存在-20°C。 避免反复冻融循环。
1.1制备 SoNa? 520 储备溶液
将 100 μL DMSO(组分 D)添加到 SoNa 520 小瓶(组分 A)中。
注意:制作一份未使用的 SoNa 520 储备溶液等份并储存在 ≤ -20 oC 下。避光并避免重复冻融循环。
2.标准溶液配制
氯化钠标准品配制
将 300 μL 蒸馏水添加到 NaCl 标准样品瓶(组分 C)中,制成 250 mM 标准储备溶液。接下来,使用测定缓冲液(组分 B)稀释该 250 mM 储备溶液以制备 40 mM (SS1)。然后进行 1:2 连续稀释以获得连续稀释的 NaCl 标准品 (SS2 – SS7)。
3.工作溶液配制
准备 SoNa? 520 工作溶液
将 100 μL SoNa 520(组分 A)添加到 5 mL 测定缓冲液(组分 B)中。用箔纸覆盖工作溶液或将其置于黑暗中,以保护工作溶液免受光照。
注意:为了获得zui佳效果,该溶液应在制备后几小时内使用。
注:5 mL 工作溶液足以进行 100 次测试。

1.根据表 1 和 2 中提供的布局准备 NaCl 标准品 (SS)、空白对照 (BL) 和测试样品 (TS)。对于 384 孔板,每孔使用 25 μL 试剂,而不是 50 μL。
2.将 50 μL SoNa 520 工作溶液添加到 NaCl 标准品、空白对照和测试样品的每个孔中,使测定体积为 100 μL/孔。对于 384 孔板,请在每个孔中添加 25 μL,总体积为 50 μL/孔。
3.将反应物在室温下避光孵育 5 至 10 分钟。
4.使用荧光酶标仪在 Ex/Em = 490/525 nm(cutoff=515nm)处检测荧光增加。
数据分析
从空白标准孔获得的读数(RFU)用作阴性对照。 从其他标准的读数中减去该值,以获得基线校正值。 然后,绘制标准读数以获得标准曲线和方程。
