Firefly Luciferase is widely used as a reporter gene to study gene regulation and function and drug screening. Since there is no endogenous Luciferase activity in mammalian cells or tissues, it is a very sensitive genetic reporter gene. Firefly Luciferase is a 62000 Dalton protein, which is active as a monomer, and does not require subsequent processing of its activity. This enzyme catalyzes the oxidation of ATP dependent D-luciferin to oxidized luciferin, producing light emission. The firefly Luciferase follows the Michaelis Menten kinetics, so the maximum light output can be achieved only when the substrate and cofactor are in large excess. When analyzing under these conditions, the light emitted by the reaction is proportional to the number of Luciferase molecules.
Note: This is a flash type luminescence detection that requires the signal to be measured immediately after adding the working solution to the sample. It requires a single tube luminescent meter or luminescent enzyme reader with a reagent syringe. Biotium also offers the Steady Luc HTS Firefly detection kit (product number 30028L), which is a homogeneous luminescent detection with a signal half-life of 3-5 hours.
Easy to use for consistent results
The firefly Luciferase detection kit is designed to simply and effectively quantify the activity of firefly Luciferase reporter enzyme in cultured cells, with high sensitivity and linearity. This kit is a new improved substitute for the Firefly Luciferase Test Kit (Catalog No. 30003). It adopts Firefly Assay Buffer 2.0, which has better working solution stability compared to the original detection buffer.
Other commercial Luciferase tests require you to dilute all firefly substrates in the test buffer, resulting in a limited shelf life after first use. In Biotium's reagent kit, the substrate is provided as a separate stock solution, which is freshly diluted for each test to achieve excellent shelf life and provide consistent signals over time.
The measurement size is based on the 96 hole format. This kit includes sufficient cell lysis buffer to perform the specified number of measurements within the size range of 96 well to 24 well plates.

Due to the lack of endogenous activity in mammalian cells or tissues, it is a highly sensitive genetic reporter gene. However, the light generated by the reaction leads to the formation of suicidal adenosyloxyfluorescein on the surface of the enzyme. It results in a very short half-life of light emission with flashing dynamics. Because of this, flash type luminescence must be detected immediately after the reaction begins, which is incompatible with batch analysis of samples.
Steady Luc from Biotium ™ The Firefly HTS detection kit contains a proprietary mixture of substances that can prevent adenosine oxyfluorescein reactions and generate persistent luminescent signals. It generates a luminescent signal with a half-life of approximately 3 hours for high-throughput microplate detection.
Compared to flash type determination, Steady Luc ™ Isoluminescent Luciferase assay has low luminescence signal. The sensitivity and detection limit of this determination will depend on the expression level of Luciferase in your experimental system and the sensitivity of the photometer.

SGVB++can be used to detect C1 in purified C1 complex (product number A098) and normal human serum samples.
