Information

Normal serums are lipid extracted to improve clarity, dialyzed against phosphate buffered saline (PBS) containing sodium azide, and freeze dried. Normal serum diluted to 5% (v/v) in PBS is strongly recommended as a blocking reagent to reduce background from non-specific, conserved sequence and/or Fc receptor binding. Best results are obtained with diluted normal serum from the same host as the labeled antibody, as a separate incubation step before addition of the primary antibody.

 

Usage

Physical State: Freeze-dried solid

Storage and Rehydration: Store freeze-dried solid at 2-8°C. Rehydrate with the indicated volume of dH2O (see product specification sheet) and centrifuge if not clear. Prepare working dilution on day of use. Product is stable for about 6 weeks at 2-8°C as an undiluted liquid.

Extended Storage after Rehydration: Aliquot and freeze undiluted product at -20°C or below. Avoid repeated freezing and thawing.

Expiration date: one year from date of rehydration. The expiration date may be extended if test results are acceptable for the intended use.

Preservative: 0.05% Sodium Azide

Suggested Working Concentration or Dilution Range:

For blocking purposes, use 5% (v/v) solution (1:20 dilution from rehydrated volume). Rehydration according to instruction yields 100% serum.

Information

Based on immunoelectrophoresis and/or ELISA, the antibody reacts with whole molecule mouse IgG. It also reacts with the light chains of other mouse immunoglobulins. No antibody was detected against non-immunoglobulin serum proteins. The antibody may cross-react with immunoglobulins from other species.

 

Whole IgG antibodies are isolated as intact molecules from antisera by immunoaffinity chromatography. They have an Fc portion and two antigen binding Fab portions joined together by disulfide bonds and therefore they are divalent. The average molecular weight is reported to be about 160 kDa. The whole IgG form of antibodies is suitable for the majority of immunodetection procedures and is the most cost effective.

 

Usage

Physical State: Freeze-dried solid

Storage and Rehydration: Store freeze-dried solid at 2-8°C. Rehydrate with the indicated volume of dH2O (see product specification sheet) and centrifuge if not clear. Prepare working dilution on day of use. Product is stable for about 6 weeks at 2-8°C as an undiluted liquid.

Preservative: None (Warning: Use of sodium azide as a preservative will substantially inhibit the enzyme activity of horseradish peroxidase.)

Suggested Working Concentration or Dilution Range:

1:500 - 1:5,000 for immunohisto/cytochemistry

1:5,000 - 1:100,000 for ELISA and Western blotting with chromogenic substrates

1:10,000 - 1:200,000 for Western blotting with ECL substrates

Dilution factors are presented in the form of a range because the optimal dilution is a function of many factors, such as antigen density, permeability, etc. The actual dilution used must be determined empirically.

PerCP (Peridinin-chlorophyll-protein complex) is isolated from Dinophyceae sp. It has an extremely high extinction coefficient, a high quantum efficiency and a large Stokes shift. It is well excited with the Argon laser at 488 nm with its maximum emission peak at 677nm. PerCP protein is commonly used for fluorescent immunolabeling, particularly in applications involving fluorescent-activated cell sorting (FACS). Its cyanine tandem conjugates (such as PerCP-Cy5.5 developed by BD) can be excited with a standard 488 nm laser and emits in the far red at a longer wavelength for multicolor flow cytometric analysis of cells. These multiple emission wavelengths make PerCP- Cyanine conjugates potentially useful fluorochromes for multicolor analysis with FITC, PE and other fluorochromes. PerCP tandem structure may make it more photostable than PerCP alone, which generally photobleaches rapidly with more powerful water-cooled gas lasers. AAT Bioquest offers iFluor protein labeling dyes that are generally brighter and more photostable than the corresponding cyanine dyes of similar wavelengths. iFluor protein labeling dyes are superior alternatives to the cyanine dyes.

图:来自拟南芥(1),大麦(2),玉米(3),烟草质膜部分的20μg总蛋白,2.5μg4) ,用蛋白质提取缓冲液,PEBAS08 300,用35体积的均质缓冲液),在70°C下变性10分钟,并在4-12%NuPageInvitrogenLDS-PAGE上分离,并印迹1hPVDF。在20mm Tris137mm氯化钠ph7.6,0.1%v/v)的封闭试剂中转移后立即封闭斑点Tween-20TBS-T)在室温下搅拌1小时。Blots在一级抗体中以1:5000的稀释度在室温下孵育1h温度搅拌。倾析出抗体溶液并将印迹短暂冲洗两次,然后洗涤一次15分钟和3次在室温下在TBS-T中搅拌5分钟。用二级抗体(抗兔IgG辣根过氧化物酶)孵育小鼠偶联的推荐二抗AS09 602)在2%封闭溶液中于室温搅拌稀释至1:20 000,持续1h。如上洗涤印迹,并根据制造商的说明用化学发光检测试剂显色5分钟。使用CCD成像仪(FluorSMaxBio-Rad)和Quantity One软件(Bio-Rad)获得印迹的图像。曝光时间为2分钟。

细胞膜荧光探针DiRAAT-22070)英文说明:

DiI, DiO, DiD and DiR dyes are a family of lipophilic fluorescent stains for labeling membranes and other hydrophobic structures. The fluorescence of these environment-sensitive dyes is greatly enhanced when incorporated into membranes or bound to lipophilic biomolecules such as proteins although they are weakly fluorescent in water. They have high extinction coefficients, polarity-dependent fluorescence and short excited-state lifetimes. Once applied to cells, these dyes diffuse laterally within the cellular plasma membranes, resulting in even staining of the entire cell at their optimal concentrations. The distinct fluorescence colors of DiI (orange fluorescence), DiO (green fluorescence), DiD (red fluorescence) and DiR (deep red fluorescent) provide a convenient tool for multicolor imaging and flow cytometric analysis of live cells. DiO and DiI can be used with standard FITC and TRITC filters respectively. Among them DiD is well excited by the 633 nm He-Ne laser, and has much longer excitation and emission wavelengths than those of DiI, providing a valuable alternative for labeling cells and tissues that have significant intrinsic fluorescence. DiR might be useful for in vivo imaging or tracing due to the effective transmission of infrared light through cells and tissues and low level of autofluorescence in the infrared range.
