流式细胞术（Flow Cytometry，FCM）是一种对液流中排成单列的细胞或其它生物微粒（如微球，细菌，小型模式生物等）逐个进行快速定量分析和分选的技术。
一、为什么要用同型对照？
同型对照是使用与一抗相同种属来源、相同亚型、相同剂量和相同的免疫球蛋白及亚型的免疫球蛋白，用于消除由于抗体非特异性与细胞结合而产生的背景染色。同型对照是真正意思上的阴性对照，它不但可以用来设定流式细胞仪的电压，而且还可以帮忙省去昂贵与繁琐的重组细胞因子竞争封闭步骤。
二、如何选择同型对照？
分析流式结果时，不能因为得到了荧光信号就判断细胞表面肯定结合有相应的荧光素，从而得出细胞表达相应的抗原分子的结论，而应该比较该荧光信号与细胞的非特异性荧光，如果得到的荧光信号大于非特异性荧光，说明得到的荧光信号部分来源于荧光素的特异性荧光，就可以得出细胞表达相应的抗原分子的结论。
所以，确定与荧光素无关的细胞自身的非特异性类光的强弱非常重要，而设立阴性对照就是为了确定细胞的非特异性荧光。
看看以下3类艾美捷整理的阴性对照流式分析比较全面的阴性对照：
第1类“negative”表示的是不加任何荧光素偶联抗体时得到的荧光信号，因为没有标记任何类光素，所以这组得到的荧光信号与荧光素无关，与细胞抗原分子CD69是否表达无关，得到的荧光信号代表的是非特异性荧光信号；
第2类“抗CD69”表示的是用抗CD69抗体标记样品细胞后得到的类光信号，虽然抗CD69抗体能修与细胞表面的CD69抗原分子结合，但是抗CD9抗体没有偶联荧光素，所以得到的荧光信号也与细胞表面是否表达有CD69分子无关，得到的荧光信号代表的也只是细胞本身的非特异性荧光信号，这种对照用于排除抗体的干扰；
第3类“IgG1-PE”表示的是标记与抗CD69抗体同种属（来源于同一物种）且同类（抗CD69-PE中的抗体IgG1类的）的非特异性抗体（IgG1）与PE荧光素偶联的同型（isotype）对照抗体（IgG1-PE）后，得到的荧光信号，这种就属于同型对照，其不能与细胞表面的CD69发生特异性结合，所以细胞表面不会结合有IgG1-PE，最后得到的荧光信号是代表细胞本身的非特异性荧光，不属于PE荧光素产生的特异性荧光信号，这种同型对照用于排除荧光素的干扰。
以上3种阴性对照，并不用全部设置齐全，一般只需要设置1种阴性对照就可以，其中第3种同型对照是最常规的阴性对照设置法。
